高浓度玻璃化冷冻剂对卵子冷冻复苏及发育潜能的影响
Effects of High Concentration of Vitrification Cryoprotectant on Revitalization and Developmental Potentiality of Human Oocytes
【摘要】 目的:探讨卵子暴露于高浓度玻璃化冷冻剂中的时间对卵子冷冻复苏及其后续发育潜能的影响。方法:收集79例未受精的成熟卵子290枚进行玻璃化冷冻及解冻,根据卵子在高浓度玻璃化冷冻剂中暴露的时间将其分为A组(147枚卵子,暴露时间10~20 s)及B组(143枚卵子,暴露时间40~50 s),比较两组卵子复苏率、受精率、卵裂率及优质胚胎率。结果:A组卵子复苏率明显高于B组(74.83%vs 63.64%,P<0.05),而两组受精率、卵裂率、优质胚胎率差异均无统计学意义(69.09%vs 68.13%;71.05%vs 61.29%;22.78%vs31.57%,P>0.05)。结论:缩短卵子在高浓度玻璃化冷冻剂中暴露的时间有助于卵子的冷冻复苏。
【Abstract】 Objective: To explore the effect of the duration that oocytes exposed to high concentration of vitrification cryoprotectants on thier revitalization and developmental potentiality.Methods: Two hundred and ninety unfertilized mature oocytes from 79 patients who accepted in vitro fertilization(IVF) embryo transplantation treatment during January 2010 to May 2011 were freezed and defrosted.The oocytes were divided into two groups according to their time exposed to cryoprotectants: group A with 147 oocytes and exposing time ranged 10 seconds to 20 seconds;group B with 143 oocytes and exposing time ranged 40 seconds to 50 seconds.Revival rates,fertility rates,cleavage rates,and high quality embryo rates of the two groups were compared.Results: Revival rate in group A was significantly higher than that in group B(74.83% vs 63.64%,P<0.05).No obvious difference was found in fertilization rate,cleavage rate or high-quality embryo rate between the two groups(69.09% vs 68.13%,71.05% vs 61.29% and 22.78% vs 31.57%,P>0.05).Conclusions: Shortening the exposure time of oocytes to high concentration of vitrification cryoprotectants is helpful to revitalization of oocytes.
