菊糖芽孢乳杆菌DS 2-18中试规模的D-乳酸发酵研究
D-lactic acid production on pilot scale by Sporolactobacillus inulinus DS2-18
【摘要】 研究了菊糖芽孢乳杆菌DS2的突变株DS2-18在中试规模的D-乳酸发酵。在容积为300L自控发酵罐中,DS2-18菌在合适的发酵条件下,即培养基组成(g/L):葡萄糖120,玉米浆8,蛋白胨6,豆粕水解液100,接种量8%(v/v),发酵温度40℃,以轻质碳酸钙作为中和剂调pH 5~6,发酵期间交替不通气和通气,发酵68 h,产D-乳酸109.4 g/L,转化率93.5%,产率1.4 g/L·h,D-乳酸的光学纯度为99.0%。研究结果还显示,豆粕水解液可以代替昂贵的酵母膏,降低D-乳酸的生产成本。本研究结果为发酵法生产D-乳酸的产业化打下了基础。
【Abstract】 D-lactic acid production on pilot scale by DS2-18,a mutant of Sporolactobacillus inulinus DS2,was investigated.Its fermentation was carried out in a 300 L automatic control fermentor.The medium were as follows(g/ L):glucose 120,corn steep liquor 8,peptone 6,soybean meal hydrolysate(SMH) 100.Under the proper conditions such as inoculum size 8%(v/v),fermentation temperature 40℃,pH 5~6 with light calcium carbonate as neutralizer, alternative aeration or no aeration in the course of fermentation duration,the concentration of D-lactic acid,the yield and productivity were 109.4%,93.5%and 1.60 g/L·h,respectively,and the optical purity of D-lactic acid was 99.0%. Meanwhile,SMH,instead of expensive yeast extracts,could reduce the manufacturing cost for producing D-lactic acid. As a result,it laid a foundation of mass production of D-lactic acid.
