葡萄藻冷冻保藏的研究

【摘要】 为给微藻大规模培养生产生物燃料提供稳定可靠的种质资源,本研究以葡萄藻为研究对象,建立了一套葡萄藻快速高效冷冻保藏的方法。通过对不同冷冻保护剂二甲基亚砜(DMSO)、甲醇(MeOH)、乙二醇(EG)、丙二醇(PG)和甘油(Gly)的毒性测试和冷冻保藏效果的比较,结果表明在以6%MeOH作为冷冻保护剂的条件下葡萄藻的存活率达到最优值98.9%。不同降温方式研究表明,程序降温对葡萄藻的冷冻保藏至关重要。葡萄藻培养时间对其冷冻保藏效果影响显著,选择对数生长阶段后期的藻体进行冷冻保藏对有利于保持较高的存活率。

【Abstract】 In order to provide stable and reliable microalgal source for the biofuel production from large scale microalgae culture,cryopreservation of Botryococcus braunii was investigated and a fast and effective method of cryopreservation was established.Five cryoprotectants(dimethyl sulphoxide,DMSO;methanol,MeOH;ethylene glycol,EG;propylene glycol,PG;and glycerol,Gly)at different concentrations were evaluated to determine their toxicity and post-thaw viability for B.braunii.The results showed that the optimal post-thaw viability of 98.9% was achieved at 6% of MeOH.Different cooling procedures were also studied and it indicated that the programmed cooling was essential for cryopreservation of B.braunii.Culture age of B.braunii affected its post-thaw viability significantly.It was suggested that the microalgae harvested from late exponential phase of culture was beneficial for cryopreservation of B.braunii.
