康复训练对脑梗死大鼠梗死灶周围SDF-1表达的影响

【摘要】 目的:研究康复训练对大鼠脑梗死灶周围基质细胞衍生因子(SDF-1)表达的影响。方法:采用线栓法制作大鼠左侧大脑中动脉栓塞(MCAO)再灌注模型。将48只造模成功成年SD大鼠随机分成造模对照组(n=24)及康复训练组(n=24)。另取24只大鼠作为假手术组。康复训练组于术后48h每天予以转棒、平衡木、滚筒等训练1h;造模对照组和假手术组则置于普通笼内饲养,不予以任何针对性训练。每组随机分为第3天、第7天、第14天、第21天4个时间点,每个时间点各取6只大鼠,分别于相应时间点进行神经行为学和运动功能评分后取脑,采用免疫组织化学方法观察各个时间点脑梗死灶周围SDF-1的表达。结果:造模术后第7天,康复训练组神经行为学评分及运动功能评分均低于造模对照组(P<0.05),神经功能明显改善;免疫组织化学染色检测,康复训练组与造模对照组大鼠脑梗死灶周围SDF-1阳性细胞于造模术后第3天增加,第7天达到高峰,并维持到第14天,第21天下降,且康复训练组在各个时间点较造模对照组增多,与第7天差异明显(P<0.05)。结论:康复训练可促进大鼠梗死灶周围SDF-1表达增加,改善神经功能。

【Abstract】 Objective:The aim of this study was to investigate the effects of rehabilitation training on the expression of stromal cell-derived factor-1(SDF-1) in the peri-infarction of focal cerebral infarction in rat model.Methods:Seventy-two male Sprague-dawley(SD) rats were tested and the peri-infarction of cerebral was induced with left middle cerebral artery occlusion(MCAO) method.Rats were randomly divided into rehabilitation training group(n=24),which was treated with bar rotating,balance beam and rolling cage exercises for 60 minutes daily at 48 hours post MCAO;control group(without training,n=24);and sham operation group(n=24).Days 3,7,14 and 21 were selected as checking timepoint.Neurological behavioral score and motor function assay were assessed among three groups.Expression of SDF-1 was done with immunohistochemistry method to account the positive cell number around the infarction spot.Results:Comparing with control and sham groups,neurological behavior score of treated group was lower(P<0.05).At days 3,7 and 14,the positive cell number in rehabilitation training group was higher than that in control and sham groups(P<0.05).Conclusion:Our data indicate that the rehabilitation training can increase the expression of SDF-1 in the infarction area.Rehabilitation may play an important role in recovering and self repairing post infarction.Detail mechanism still needs further study.
