重复性“饥饿/再投喂”对大鼠脂代谢及主动脉影响的研究
The Influence Research of Repeated Fasting/Refeeding on Lipid Metabolism and Aorta in Rats
【摘要】 为探讨经过重复性"饥饿/再投喂"方式处理后大鼠摄取高脂食物时有关体质量控制与能量平衡调节的瘦素(leptin)和神经肽Y(NPY)的表达变化,设计"重复性饥饿1 d/再投喂基础饲料1 d,持续6周"的饲喂方式处理动物模型,6周后改为每天喂以高脂饲料(高脂模型组),另设高脂对照组和基础对照组,喂养至12周,测定了血脂水平和血清leptin含量,RT-PCR法检测了主动脉神经肽Y(NPY)mRNA的表达。结果表明,模型组血清leptin水平显著高于对照组(P<0.05),主动脉NPY mRNA表达水平则显著低于对照组(P<0.01);高脂模型组血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)及leptin水平均显著低于高脂对照组而高于基础对照组(P<0.05),主动脉NPY mRNA表达水平显著高于其他两组(P<0.01),主动脉病变程度则相对较轻。提示重复性"饥饿/再投喂"饲喂处理可能激活了leptin受体调节能量平衡以控制体质量这一调节系统,可减轻高脂饮食诱导的leptin抵抗,NPY可能参与了其对能量平衡的调节过程。
【Abstract】 To explore the expression changes of leptin and NPY associated with body weight control and energy balance regulation after repeated fasting/refeeding rats given high fat diet,a model of repeating fasting/refeeding rats(repeated cycles of 1 d fasting and 1 d refeeding for 6 week fed with common fat diet)was designed,then the model group was switched to high fat diet everyday(high-fat model group),and the control group was divided into high-fat and common-fat control groups.By the end of 12th week,the concentrations of serum lipid and leptin were measured,the expression of NPY mRNA in aorta were detected by RT-PCR.The results showed that the serum leptin level of model group was significantly higher than that of control group(P<0.05),while the expression of NPY mRNA in aorta was significantly lower than that of control group(P<0.01);The serum TC,TG,LDL-C,and leptin levels of high-fat model group were significantly lower than those of high-fat control group whereas significantly higher than those of common-fat control group(P<0.05),while the expression of NPY mRNA in aorta was significantly higher than that of high-fat and common-fat control groups(P<0.01),the lesion degrees of aorta were also lighter relatively.It concluded repeated fasting/refeeding feeding pattern may the leptin system of energy balance to control body weight,can reduce the leptin resistance induced by high fat diet,NPY may the process of energy balance regulation.
