氟化钠对小鼠的急性毒性实验研究
Experimental Study on Mice’s Acute Toxicity of Sodium Fluoride
【摘要】 为探讨氟化钠对小鼠的急性毒性及反应,采取灌胃法给小鼠一次性给药,观察了7 d内35.00、55.56、88.18、139.97、222.18、352.67、559.79 mg/kg等7个剂量组小鼠的毒性反应和死亡情况,计算了氟化钠经口半数致死量(LD50)及95%可信限。结果表明,各剂量组中毒小鼠均出现不同程度的抽搐、呼吸心跳加快等症状;氟化钠经口LD50为168.34 mg/kg,95%可信限为125.89~225.13 mg/kg,属于中等毒性;在一定范围内,小鼠死亡率与氟化钠染毒剂量成正相关(r=0.996,P<0.001);大体解剖可见胃扩张、膨大,并伴有明显的出血、淤血表现,同时小鼠的肾脏、肝脏、小肠等组织器官有广泛性损伤,并有不同程度的出血、淤血等病理变化。
【Abstract】 To study and determine the acute toxicity of sodium fluoride on mice,mice were given sodium fluoride via gastric tube gavage through as one-time administration at doses of 35.00,55.56,88.18,139.97,222.18,352.67,559.79 mg/kg respectively.Mice were observed for 7 days after sodium fluoride.The median lethal does of oral sodium fluoride(LD50)and 95% confidence interval were calculated.The results showed that all mice in each dose group present different levels of seizures,respiratory symptoms and tachycardia.The oral sodium fluoride oral LD50=168.34 mg/kg,with 95% confidence interval 125.89~225.13 mg/kg,which is moderate toxic.Within a certain range,mortality and sodium fluoride dose in mice was positively correlated(r=0.996,P<0.001).The autopsy revealed visible gastric edema and dilatation,accompanied by significant bleeding and congestion.In the meantime,the kidneys,liver,small intestine and other organs have extensive damage with varying degrees of hemorrhage,congestion and other pathological changes.
