远志总皂苷对神经干细胞向神经元分化的影响

【摘要】 目的:观察远志总皂苷对神经干细胞(NSCs)向神经元分化的影响。方法:分离、培养并鉴定新生大鼠海马区NSCs,分别添加终浓度5μg/mL、20μg/mL及100μg/mL的远志总皂苷进行诱导1 d或3 d。免疫细胞化学法检测神经元特异性烯醇化酶(NSE)阳性细胞。结果:原代与传代培养的细胞均可持续分裂增殖形成悬浮生长的神经球,经远志总皂苷诱导后1 d、3 d,NSE阳性神经元数量较未接受诱导的细胞明显升高(P<0.01)。结论:远志总皂苷对海马NSCs向神经元分化有一定的促进作用。

【Abstract】 Objective: To observe the effects of polygalaceae saponins(TEN) on neural stem cells(NSCs) differentiation into neurons.Methods: NSCs were isolated from rat hippocampus,cultured and identified.TEN with a final concentration of 5 μg/mL,20 μg/mL,and 100 μg/mL was respectively used to induce differentiation of NSCs.The expression of neuron specific enolase(NSE) was detected by cell immunocytochemistry at days 1 and 3 after induction.Results: Primary and sub-cultured cells proliferated to form neurospheres.After treatment by TEN for 1 or 3 days,the expression of NSE was significantly increased(P<0.01).Conclusion: TEN can enhence the differentiation of the hippocampal NSCs into neurons.
