pEGFP/A2M(FP6)重组质粒转染小鼠胚胎神经干细胞的实验研究
Transfection of pEGFP/A2M(FP6) Plasmid Vectors into Mouse Embryonic Neural Stem Cells
【摘要】 目的:观察携带pEGFP/A2M(FP6)重组质粒的神经干细胞(NSCs)的生物学特性。方法:培养NSCs,采用Nucleofector转染方法将pEGFP/A2M(FP6)重组质粒转染至NSCs,利用其携带的绿色荧光和RT-PCR观察其转染情况,免疫组织化学法检测A2M(FP6)蛋白表达。结果:Nucleofector转染方法可成功高效地将pEGFP/A2M(FP6)重组质粒转染至NSCs,且对NSCs的增殖、分化无明显影响,转染细胞出现A2M(FP6)的表达。结论:携带pEGFP/A2M(FP6)质粒的NSCs仍具有增殖、分化特性,并具有合成A2M(FP6)的能力。
【Abstract】 Objective:To study the biological features of the mouse embryonic neural stem cells(NSCs) transfected by pEGFP/A2M(FP6).Methods:NSCs of mouse embryos were isolated and cultured.The pEGFP/A2M(FP6) plasmid vectors were transferred into the cultured NSCs by Nucleofector TM.The expression of A2M FP6 mRNA was detected with RT-PCR and immunofluorenscent analysis.Results:The pEGFP/A2M(FP6) plasmid vectors were successfully transferred into NSCs in which expression of A2M(FP6) remained rich.In addition,immunohischemistry showed that the proliferation and differentiation of NSCs were unchanged.Conclusion:The proliferation and differentiation of NSCs after tansfection with pEGFP/A2M(FP6) were unaltered,keeping the ability to produce A2M(FP6).
