藏药柳茶提取物对非酒精性脂肪肝病大鼠肝脏保护作用及其机制研究

【摘要】 目的研究藏药柳茶提取物(治疗胃胀痛及消化不良藏药)对非酒精性脂肪肝病(NAFLD)大鼠肝脏保护作用的机制。方法用高脂饮食复制NAFLD大鼠模型,随机分为4组:模型组、柳茶提取物低、中、高剂量组;同时设立正常对照组,相应干预8周后,处死。制作肝组织切片观察形态变化;检测血清ALT、AST水平;荧光定量RT-PCR法测抗氧化酶GSH-PX2及CAT mRNA表达量。结果与模型组相比,柳茶干预组肝细胞损伤减轻,肝功能改善,CAT mRNA表达显著增加,GSH-PX2 mRNA表达有不同程度增加。结论藏药柳茶提取物对NAFLD大鼠肝脏损伤具有保护作用;可能与上调肝脏抗氧化酶GSH-PX2、CATmRNA的表达,增强其抗脂质过氧化能力有关。

【Abstract】 Objective To investigate the protective mechanism of Liucha extract against non-alcoholic fatty liver disease(NAFLD)in rats.Methods NAFLD models of SD rats were established by high fat diet feeding for 6 weeks,and then divided randomly into four groups: the model group,low-,medium-and high-dosage Liucha extract treatment groups.A normal control group was also run.Eight weeks later,all rats were killed.Hepatic morphological changes were observed under light microscope and liver function parameters(ALT,AST) were examined.Expression of glutathione peroxidase 2(GSH-PX2),catalase(CAT) mRNA were measured by fluorescent quantitation RT-PCR.Results Compared with model group,hepatic cell damage were palliated,liver function were improved and the expression of CAT mRNA was significantly increased in Liucha extract treatment groups.GSH-PX2 mRNA was increased in different level.Conclusion Liucha extract has a good protective effects on NAFLD in rats,its mechanism is probably related to up-regulate the expression of liver antioxidase GSH-PX2,CAT mRNA and enhance the action of hepatic anti-lipidperoxidation.
