不同浓度血管内皮细胞生长因子皮下局部注射对大鼠脂肪移植体存活率的影响
Effect of subcutaneous local injections of different concentrations of vascular endothelial growth factors on the survial rate of autologous fat grafts in rats
【摘要】 目的:应用外源性血管内皮生长因子(VEGF)局部注射移植颗粒脂肪组织的方法,观察VEGF对移植组织早期的血管增生情况以及提高脂肪存活率的效果。方法:成年雌性Wistar大鼠随机分3组:生理盐水注射组、VEGF 100ng注射组、VEGF 200ng注射组。于术后第10,30天取出各组的脂肪移植体,HE染色后,光镜下观察各组移植体脂肪细胞形态学的改变;免疫组化法进行微血管密度的定量,测量移植体第30天的剩余体积。结果:实验组的移植物保存体积和微血管密度明显高于生理盐水对照组,有显著性意义(P<0.05),VEGF组组间差别无统计学意义(P>0.05),实验组脂肪成活率高于对照组。结论:有效浓度的VEGF能够提高移植后颗粒脂肪的存活率。
【Abstract】 Objective: Exogenous vascular endothelial growth factors(VEGF) local injections of transplanted particles of adipose tissue was adopted in the experiment,and then the early period of angiogensesis conditions in the recipient site and the effect of improving the survival rate of fat cells were observed.Methods: Wistar rats weighing 250 g to 280 g were randomly divided into three groups: sterile saline injection group(control group).100 ngml VEGF injection group,and 200 ngml VEGF injection group.The transplanted fat in each group was taken out respectively on the 10th and 30th day after surgeries.The morphological changes of the fat cells in each group were observed after the transplanted fat were stained with hematoxylin-eosin(HE).Also,immunohistochemistry method was applied to quantifying the microvessel-density and determing the remained volumes of grafts on the 30th day.The data of microvascular numbers and sizes were processed by SPSS.Results: The remained volume of the grafts and the microvascular density in the experimental groups were obviously higher than those in the control group,which were of statistical differences(P<0.05),but no obvious difference between the VEGF groups(P>0.05).In addition,the survival rate of fat in the experimental groups was higher than that in the control group.Conclusions: The effective concentrations of VEGF can improve the survival rate of fat particles in rats after transplantation.
