半边旗提取物5F下调炎症因子表达对荷S180肉瘤小鼠的抗肿瘤活性的影响

【摘要】 目的观察半边旗提取物5F对荷S180肉瘤小鼠的抗肿瘤作用及炎症因子影响。方法建立荷S180肉瘤腹水瘤及实体瘤小鼠模型,随机分为生理盐水组、5-Fu组及低、中、高剂量5F组;采用腹腔注射给药,末次给药后处死小鼠,记录腹水瘤小鼠的生存时间,检测实体瘤小鼠的抑瘤率、白细胞数、胸腺指数、脾指数及血清IL-1、IL-6、TNF-α和瘤内TNF-α水平。结果低、中、高剂量5F组小鼠的中位生存时间分别为(15.4±0.5)、(21.4±1.3)和(28.2±0.7)d,明显高于生理盐水组(P<0.05)。5F组瘤重明显低于生理盐水组(P<0.05);中、高剂量5F对胸腺指数、脾指数、IL-1、IL-6及TNF-α水平有明显抑制作用(P<0.05),但对白细胞数无明显影响。结论 5F可有效抑制小鼠荷S180肉瘤的生长,可能与下调炎症因子表达有关。

【Abstract】 Objective To observe the role of 5F from Pteris semipinnata L.in antitumor activity and inflammatory cytokines S180 sarcoma-bearing mice.Methods Mouse models with S180 ascites and solid sarcoma were divided into normal saline(NS),5-Fu,and low,middle and high dose 5F groups.The mice were sacrificed after the last peritoneal administration.The survival time was recorded in ascites sarcoma mice,while the inhibitory rate,WBC,thymus index(TI),spleen index(SI),serum levels of IL-1,IL-6 and TNF-α,and intratumoral content of TNF-α were detected in solid sarcoma mice.Results The median survival time of low,middle and high dose 5F groups were(15.4±0.48),(21.4±1.28),and(28.2±0.73) days,respectively,and they were superior to those in NS group(P<0.05).Tumor weight in 5F groups was significantly lower than that in NS group(P<0.05).Compared with NS group,TI,SI,and levels of IL-1,IL-6 and TNF-α decreased(P<0.05),but WBC showed no change in middle and high dose 5F groups.Conclusion 5F can inhibit the growth of S180 transplanted-tumor in mice,which might be related to down-regulation of inflammatory cytokines.
