三氧化二砷对瘢痕成纤维细胞增殖与胶原合成的影响
Effect of arsenic trioxide on proliferation and collagen synthesis of scar-derived fibroblasts
【摘要】 目的研究三氧化二砷对瘢痕成纤维细胞增殖和胶原合成的影响。方法以四甲基偶氮唑盐(MTT)方法和吸光度检测法及逆转录聚合酶链反应(RT-PCR)法测定三氧化二砷对成纤维细胞增殖和培养基中的胶原含量以及I、III型前胶原mRNA表达的影响。结果三氧化二砷对成纤维细胞的增殖具有显著的抑制作用,还能显著降低培养基中可溶性胶原的含量(P<0.01)。上述抑制作用呈现药物浓度和作用时间依赖性。采用RT-PCR法对I、III型前胶原的mRNA检测显示随着药物浓度增高前两者的表达量降低。结论氧化二砷对瘢痕成纤维细胞的增殖和胶原合成具有显著的抑制作用。
【Abstract】 Objective To study the effect of arsenic trioxide on proliferation and collagen synthesis of scar-derived fibroblasts.Methods After treated with arsenic trioxide,the proliferation,collagen content,and type I and III procollagen mRNA of scar-derived fibroblasts were determined by MTT,photometry and RT-PCR,respectively.Results Arsenic trioxide treatment resulted in dose-and time-dependent inhibition of fibroblast proliferation and collagen production(P<0.01) and dosedependent downregulation of type I and III procollagen expression.Conclusion Arsenic trioxide can inhibit the proliferation and collagen synthesis of scar-derived fibroblasts.
