补骨胶囊对骨质疏松大鼠牙种植体结合骨板形成的影响
Effects of Bugu capsules on dental implant osseintegration in osteoporositic rats
【摘要】 目的研究中药复方制剂补骨胶囊对骨质疏松大鼠牙种植体结合骨板形成的影响。方法 8月龄雌性SD大鼠45只随机分为假手术组、卵巢切除组和中药组。术后3个月确定骨质疏松模型建立后,在大鼠胫骨近中干骺端植入纯钛螺纹柱状种植体,术后第1天开始给中药组大鼠用补骨胶囊冲剂灌胃,其它两组灌服生理盐水。术后1、2、3个月分批处死大鼠,取带种植体的胫骨行形态计量学、组织病理学和成骨蛋白(BMP-2)免疫组化检查。结果中药组结合骨板连续性较好,骨小梁周围成骨细胞较多,术后1~2个月BMP-2阳性细胞数明显高于卵巢切除组和假手术组(P<0.05)。结论骨质疏松可影响种植体的骨结合,补骨胶囊可使骨质疏松大鼠的种植体结合骨板宽度、种植体周围松质骨区骨量、成骨细胞、BMP-2表达显著增加。
【Abstract】 Objective To investigate the effects of Bugu capsules on dental implant osseintegration in osteoporositic rats.Methods Forty-five 8-month-old female SD rats were randomly divided into sham group,ovariectomized group(OVX group) and Bugu capsule group(BGC group).After 3 months,the osteoporositic models were identified,and then implanted by pure titanium screw-implants in the proximal metaphyses of tibiae.On day 1 after surgery,BGC group received oral Bugu capsules,whereas other two groups received normal saline.The rats were sacrificed at 1,2 and 3 months after surgery,the implanted tibiae were observed by morphometry,histopathology and bone morphogenetic protein(BMP-2) immunohistochemistry.Bone histomor-phometric parameters were measured to quantify the changes of new bone around titanium implants and to observe the changes of implant-bone interface.Results Compared with OVX and sham groups,bone lamella continuity was satisfactory,numerous osteoblasts were present around trabeculae,and BMP-2 positive cells increased remarkably in BGC group 1~2 months after surgery(P<0.05).Conclusion Osteoporosis can inhibit the osseointegration of titanium implant.Bugu capsules can promote the combined bone lamella width,cancellous bone mass,osteoblasts,and BMP-2 expression after titanium implant in osteoporositic rats.
