TGF-β1及CTGF在胆管缺血性损伤修复过程中的表达及意义
Expression and significance of TGF-β1 and CTGF in the repairing process of bile duct ischemic injury
【摘要】 目的观察TGF-β1及CTGF在胆管损伤修复过程中的表达,探讨良性胆管狭窄形成机制。方法建立大鼠胆管缺血损伤模型,观察术后胆总管及肝脏组织病理改变;免疫组化检测两组胆管组织TGF-β1及CTGF的表达,计算阳性细胞数。结果实验组夹闭处胆管管腔狭窄伴纤维组织增生,钳夹部位以上胆管扩张;肝脏炎性细胞浸润。对照组肝脏及胆管组织无异常。实验组胆管TGF-β1及CTGF呈高水平正相关表达(r=0.246,P<0.05),对照组表达较弱,两组比较差异有统计学意义(P<0.01);实验组组间比较差异无统计学意义(P>0.05)。结论 TGF-β1和CTGF具有协同作用,引起成纤维细胞增殖、细胞外基质过度沉积,是导致良性胆道狭窄形成的重要因素。
【Abstract】 Objective To investigate the expression of TGF-β1 and CTGF in the tissue of common bile duct during wound healing and explore their roles in the pathogenesis of benign biliary stricture.Methods A model of bile duct trauma was surgically established in mice.The samples of common bile duct and liver were extracted for microscopic examination after operation.Immunohistochemistry technique was applied to detect the distribution of TGF-β1 and CTGF in common bile duct of two groups and the quantity of positive cells was also calculated.Results Obvious dilatation of the common bile duct above the clamped site was observed,while the bile duct lumen became smaller or occluded in the clamped site accompanied with fibrosis.Lots of inflammatory cells were seen in the specimens of liver.There were no apparent pathological changes in control group.The positive expression of TGF-β1 and CTGF was remarkably stronger than those in normal bile duct,respectively(P<0.01),while no significant difference among each time point in experimental group(P>0.05).Correlation analysis revealed that there was a positive relationship between the expression of TGF-β1 and that of CTGF(r=0.246,P<0.05).Conclusion The high expression of TGF-β1 and CTGF is correlated closely with the fibroblast proliferation,extracellular matrix overdeposition in the process of benign biliary stricture.
