NF-κB抑制剂PDTC联合阿霉素对肝癌HepG2细胞增殖的影响
EFFECT OF NUCLEAR FACTOR-ΚB INHIBITOR PDTC COMBINED WITH ADRIAMYCIN ON THE HEPATOCELLULAR CARCINOMA CELL HEPG 2
【摘要】 目的探讨核转录因子-κB(NF-κB)抑制剂吡咯烷二硫氨基甲酸(PDTC)与阿霉素(ADM)联合应用对抑制HepG-2细胞增殖的效应及其机制。方法培养人肝癌HepG-2细胞株,分为空白组,ADM组、PDTC组、PDTC+ADM组,观察HepG-2细胞的生长情况,MTT比色法检测细胞增殖,利用中效原理判定药物合用的效果,免疫组化检测NFκB P65表达情况。结果 ADM及PDTC均可有效抑制细胞的生长,联合用药的中效浓度比单用时减少明显。阿霉素和PDTC两药小剂量合用时可产生协同作用CI<1。PDTC+ADM组NFκBp65表达显著低于正常肝癌细胞组(P<0.05)。结论 PDTC和ADM均可抑制肝癌细胞的生长,小剂量合用产生协同作用,其协同机制可能与PDTC抑制阿霉素对NFκB的激活有关。
【Abstract】 Objective To study the effect and possible mechanism of pyrrolidine dithiocarbamate(PDTC),a potent inhibitor of nuclear factor kappa B(NF κB),combined with adriamycin(ADM) on human hepatocellular carcinoma cell HepG-2.Methods HepG-2 cells were incubated and divided into ADM group,PDTC group,and PDTC+ADM group.The cells growth activity was measured by growth curve and the proliferation and cell apoptosis rates were evaluated by MTT reduction assay and the median-effect principle was also used.The level of NF κBP65 were detected by immunohistochemistry.Results PDTC and ADM could inhibit HepG-2 cells growth effectively which could be augmented as they combined.It can lead to the synergistic effect at lower concentration(CI<1).The expression of NF κB P65 in PDTC plus ADM group was lower than that in normal group.Conclusions Both PDTC and ADM can cause human hepatocellular carcinoma cell HepG-2 growth inhibition.They lead to the synergistic effect,whose mechanism may be associated with PDTC which can inhibit the activation of NF-kappaB activation in Adriamycin.
