产朊假丝酵母C-27利用甜高粱秸秆汁产SCP的培养基优化
Fermentation medium optimization for single cell protein production with sweet sorghum stalk juice by Candida utilis C-27
【摘要】 应用二次正交旋转组合试验设计方法对产朊假丝酵母C-27产单细胞蛋白(SCP)的发酵培养基组分(甜高粱秸秆汁、葡萄糖、蛋白胨、MgSO4和KH2PO4)进行了优化。结果显示,甜高粱秸秆汁和蛋白胨的浓度对SCP的产量有显著的正效应,而其他组分未表现出显著的影响。产朊假丝酵母C-27产SCP最终的优化发酵培养基配方为:甜高粱秸秆汁220 mL/L、葡萄糖3.25 g/L、蛋白胨37.5 g/L、MgSO40.325 g/L、KH2PO40.325 g/L。该优化的发酵培养基配方下SCP产量达8.72 g/L,比初始发酵培养基配方(甜高粱秸秆汁120 mL/L、工业葡萄糖20 g/L、工业蛋白胨20 g/L、MgSO4.7H2O 2.0 g/L、KH2PO42.0 g/L)下的SCP产量(7.00 g/L)提高了24.57%。
【Abstract】 In this paper,the fermentation medium(sweet sorghum stalk juice,glucose,peptone,MgSO4 and KH2PO4) of SCP-producing strain Candida utilis C-27 was optimized with the method of rotational quadratic orthogonal experiment design.The result showed that,sweet sorghum stalk juice and peptone had significant effect on SCP production,and the optimal fermentation medium for Candida utilis C-27 fermentation was as following,sweet sorghum stalk juice 220 mL/L,glucose 3.25 g/L,peptone 37.5 g/L,MgSO4 0.325 g/L,KH2PO4 0.325 g/L. The final SCP yield under this fermentation medium reached 8.72 g/L,which was 24.57% higher than that obtained with the original medium recipe(sweet sorghum stalk juice 120 mL/L,glucose 20 g/L,peptone 20 g/L,MgSO4 2.0 g/L,KH2PO4 2.0 g/L).
