薤白皂苷化合物对CD40L表达及血小板中性粒细胞黏附的影响
Furostanol saponins from Allium Macrostemon Bunge Bulbs inhibit Platelet CD40L expression and leukocyte-platelet adhesion
【摘要】 目的探讨薤白皂苷化合物对血小板表面CD40L表达及血小板中性粒细胞黏附的影响。方法制备富含血小板血浆和中性粒细胞,用荧光实时定量PCR和玫瑰花结计数的方法检测经不同浓度薤白化合物单体预处理后血小板表面CD40L的表达及其与中性粒细胞之间的黏附性。结果在6个薤白皂苷单体化合物中,化合物1、4、6呈剂量依赖性抑制血小板CD40L的表达,并明显抑制ADP诱导的血小板与中性粒细胞之间的黏附。结论薤白皂苷化合物可能具有抗血小板相关炎症的作用。
【Abstract】 Objective To investigate the inhibitary effects of furostanol saponins from Allium Macrostemon Bunge Bulbs on Platelet CD40L expression and polymorphonucelear leukocytes-platelet(PMN-platelet) adhesion. Methods Human platelets and neutrophils were prepared and treated with different isolated furostanol saponins at different concentrations.Expression of CD40 on ADP activated platelets and PMN-platelet adhesion activity was measured by real time PCR and rosette assay,respectively. Results Significantly inhibitary effect was observed in compound 1,4 and 6 isolated from Allium Macrostemon on the expressions of CD40L and PMN-platelet adhesive activity on ADP-induced active platelets following concentration-dependent manners(P<0.05). Conclusion The furostanol saponins from bulbs of Allium Macrostemon Bunge potentially inhibit activity on platelet inflammation.
