棉花黄萎病菌ITS序列的获得及系统进化分析

【摘要】 为探明从河南安阳棉区分离的棉花黄萎病菌的系统进化关系,利用真菌内转录间隔区通用引物ITS1和ITS4对来自安阳地区的棉花黄萎病菌内转录间隔区(ITS)进行PCR扩增,得到542bp大小的片段,经过克隆、测序和在GenBank中进行BLASTN分析,证明了该片段来自大丽轮枝菌的ITS区,也进一步证明该棉花黄萎病菌是大丽轮枝菌。同时,对该序列在NCBI的GenBank进行了登记,登记号为EU835817。利用该ITS序列与GenBank中搜索到的黄萎病菌ITS序列一起构建了相应黄萎病菌的系统进化树,在进化树上所搜索到的13个大丽轮枝菌都聚类到一个进化枝上,而且中国报道的植物黄萎病菌,包括安阳地区黄萎病菌在内的3个大丽轮枝菌在进化树上处于相邻的位置,表明:这3个菌可能具有相同的起源。这些结果有助于理解棉花黄萎病安阳菌株的进化地位,该ITS序列还可以应用于棉花黄萎病安阳菌株的特异分子鉴定。

【Abstract】 The ITS segment with 542 bp was obtained by amplifying ITS of cotton V.dahliae strain from Anyang area to understand the phylogenetic evolution relationship of cotton V.dahliae strains.The BLASTN analysis showed that the amplified segment was from ITS area of V.dahliae really.The sequence was registered in GenBank of NCBI and its register number is EU835817.The phylogenetic evolution tree of relevant V.dahliae strains was established by using the amplified ITS sequence and the ITS sequences of V.dahliae in Genbank.13 V.dahliae strains in the phylogenetic evolution tree were clustered into one evolution branch and three V.dahliae strains including Anyang V.dahliae strain reported by China were on the adjacent location of the phylogenetic evolution tree,which indicated that the three strains might be of the same origin,the result helped to understand the evolution position of Anyang V.dahliae strain and the ITS sequence(EU835817) could be applied in identification of specific molecular of Anyang V.dahliae stain.
