鸭LXRα基因cDNA的克隆及序列分析

【摘要】 为鸭肝X受体α(LXRα)基因表达、结构功能及其在鸭脂代谢信号传导中的相关研究奠定基础资料,以兴义鸭肝脏组织总RNA为模板,采用RT-PCR法对LXRα基因进行扩增测序,分析其序列特征。结果表明:鸭LXRα基因扩增产物克隆拼接的cDNA序列全长为1 412 bp(GenBank登录号:FJ966078),编码区(CDS)长1 230 bp,包含起始密码子ATG和终止密码子TGA,编码409个氨基酸残基的LXRα锌指蛋白,含有17个磷酸化位点、3个糖基化位点、2个锌指结构(P-box和D-box)和1个配体结合区(LBD),无跨膜螺旋结构,表现出典型的核激素受体超家族的结构特征。鸭LXRα蛋白与哺乳动物和鱼类的LXRα蛋白的同源性为73%~76%,与禽类的同源性高达97%左右,说明LXRα基因在系统进化过程中具有很高的保守性。

【Abstract】 The cDNA of liver X receptor alpha(LXRα) gene was cloned from total RNA of liver in Xingyi duck by RT-PCR and its sequence features were analyzed to lay the foundation for duck LXRα expression,structural function and its effect in signal transduction of lipid metabolism.The results showed that the duck LXRα cDNA was 1412bp in length(GenBank accession number: FJ966078),the open reading frame 1230bp,including the start codon ATG and stop codon TGA,and encoded a deduced LXRα protein with 409 amino acids residues.LXRα was a zinc finger proteins with 17-phosphorlated sites,3-glycosylated sites,2-zinc finger domains(P-box and D-box) and a hydrophobic ligand-binding domain(LBD) without transmenbrane helices,and displayed the common structural features of the nuclear hormone receptor superfamily.The identity of amino acids sequence of duck LXRα with mammals and fish were about 73%~76%.However,it was about 97% with that of avian.The results suggested that the LXRα gene was highly conserved in evolution.
