高效液相色谱法同时测定虫草素和腺苷
Simultaneous Determination of Cordycepin and Adenosine by High Performance Liquid Chromatography
【摘要】 本文用微波法提取蛹虫草培养基和子实体中的虫草素及腺苷,并利用高效液相色谱法同时测定虫草素和腺苷。选择Inertsil ODS-SP柱(150×4.6mm,5μm)为色谱柱,乙腈-水为流动相,流速1.0mL/min,柱温35℃,二极管阵列检测器(DAD)检测,检测波长260nm,进样量10μL。在2~10μg/mL浓度范围内,虫草素和腺苷线性关系良好,其相关系数分别为0.9998和0.9992。虫草素和腺苷的回收率分别是63.30%~67.11%和63.97%~70.39%,相对标准偏差分别是0.56%~1.97%和0.94%~1.97%,检出限分别是0.19μg/mL和0.02μg/mL。
【Abstract】 Cordycepin and adenosine from the culture medium of cordyceps militaris and cordyceps were separated by microwave extraction.Their concentrations were determined by high performance liquid chromatography.The chromatographic separation was achieved on a Inertsil ODS-SP column(150×4.6 mm,5 μm),using acetonitrile-water as the mobile phase with a flow rate of 1 mL/min at 35℃,a diode array detector(DAD) with 260 nm and a injection volume of 10 μL.The calibration curves of cordycepin and adenosine were linear over the concentration range of 2~10 μg/mL,and the correlation coefficients were 0.9998 and 0.9992.The standard recoveries were 63.30%~67.11% and 63.97%~70.39% with the RSDs of 0.56%~1.97% and 0.94%~1.97%,and the LODs were 0.19 μg/mL and 0.02 μg/mL,respectively.
