基于纳米结构双层类脂膜的雌激素传感器

【摘要】 采用铂盘电极支撑的纳米金修饰的双层类脂膜(Pt/BLM/Au)来固定雌激素受体(ER),制备Pt/BLM/Au/ER用于检测雌激素的传感器。电极不同修饰状态分别用循环伏安法和交流阻抗法进行表征。以交流阻抗法测得电极结合雌激素后阻抗的改变与17β-雌二醇(E2)的浓度在5~150ng/L范围呈良好线性关系,相关系数r=0.996,检出限(S/N=3)为1ng/L。将其用于环境样本检测,结果和MCF-7细胞增殖实验结果相关性好。

【Abstract】 There is a growing demand for new technologies that are capable of screening the wide variety of xenoestrogens in environment.Here,a nanostructure electrochemical biosensor(Pt/BLM/Au/ER) for directly screening estrogenic substances based on estrogen receptor(ER) binding was developed by immobilization of ER in bilayer lipid membrane(BLM) modified with Au nanoparticles,which was supported by the platimun disk electrode.The properties of the modified electrodes were characterized by cyclic voltammetry and impedance spectroscopy.The results indicated that the impedance change of the biosensor was linear with the concentration of natural estrogen 17β-estradiol(E2) ranging from 5 to 150 ng/L with a correlation coefficient of 0.996.And the detection limit is 1 ng/L.The biosensor was easily performed and showed good selectivity,and the Au nanoparticles greatly enhanced the sensitivity and stability of the biosensor.Moreover,estrogenic activity of water samples determined by this biosensor was in good agreement with that determined by MCF-7 cell proliferation assay.
