金纳米微粒修饰毛细管电泳分离蛋白质的研究

【摘要】 报道了金纳米微粒(Au NP)修饰毛细管电泳分离蛋白质的方法。采用物理吸附法将Au NP修饰在熔融石英毛细管内表面,制备成Au NP修饰毛细管。探讨了修饰剂Au NP的浓度对电渗流及蛋白质分离的影响。结果表明,Au NP修饰的毛细管能有效地抑制电渗流及蛋白质在毛细管内壁上的吸附,提高分离效率。在优化的实验条件下,实现了人血清白蛋白(HSA)与牛血清白蛋白(BSA)两种蛋白质的基线分离,且HSA与BSA的浓度与其峰面积呈现良好的线性关系,相关系数分别为0.9843和0.9948,检出限(S/N=3)分别为0.83μg/mL和0.80μg/mL。

【Abstract】 Gold nanoparticles were coated on the inner surface of fused silica capillaries by physical adsorbtion,and various factors influencing the electroosmotic flow and separation efficiency of two similar proteins were investigated and optimized.Compared with the unmodified capillaries,the electroosmotic flow and the adsorption of the proteins on the capillary wall was obviously decreased on the gold nanoparticles coated capillaries.Under the optimized separation conditions,HSA and BSA were separated in a 20 mmol/L borate buffer(pH=10.5) containing 1.08 nmol/L gold nanoparticles.Good linearity was obtained over the concentration range of 2.0×10-7~1.0×10-6 mol/L with correlation coefficients of 0.9943 and 0.9848,and the detection limits(S/N=3) were 0.83 and 0.80 μg/mL for HSA and BSA,respectively.
