非小细胞肺癌中端粒酶催化基团mRNA表达的临床意义及其与P53的相关性

【摘要】 目的:研究非小细胞肺癌与正常肺组织穿刺标本中端粒酶催化基团mRNA表达与肿瘤分期与分级关系,探讨其在非小细胞肺癌早期诊断中的临床意义。观察肿瘤抑制基因P53与hTRT mRNA表达的关系。方法:采用原位杂交技术检测非小细胞肺癌与正常肺组织中端粒酶催化基团mRNA表达;用免疫组化方法检测非小细胞肺癌的P53蛋白表达。结果:30例非小细胞肺癌中25例端粒酶催化基团mRNA表达阳性,阳性率83.3%;对照组20例中无端粒酶催化基团mRNA阳性表达,阳性率0%。非小细胞肺癌中,hTRT mRNA表达与P53蛋白表达呈正相关(r=0.577,P<0.01)。结论:端粒酶催化基团mRNA在非小细胞肺癌中有异常阳性表达,表达与肿瘤分期与分级有关。与肿瘤病理分型无关。P53突变可能与非小细胞肺癌hTRT基因转录激活有关。

【Abstract】 Objective To evaluate the expression of human telomerase reverse transcriptase messenger RNA(hTRT mRNA) in non small cell lung cancer tissue samples and to explore the diagnostic efficacy,and to study the effect of tumor suppressor gene P53 on the expression of hTRT mRNA.Methods We investigated the expression of hTRT in 30 cases of non small cell lung cancer.,and 20 cases lung tissues by in situ hybridization.The expression P53 protein were examined by immunohistochemistry in 30 carcinomas.Results Of 30 cases non small cell lung cancer,25(83.3%) cases had norderate expression of hTRT mRNA,while no expression hTRT mRNA in normal tissues.The expression level of hTRT mRNA was significantly different from normal tissue(P<0.01).While the expression level of hTRT mRNA was significantly different from cancer stage(P<0.05).The positive correlation between P53 and hTRT mRNA was found in non small cell lung cancer(r=0.577,P<0.01).Conclusion Non small cell lung cancer tissues have increased positive expression of hTRT mRNA,yet the expression has relationship with cancer stages and grades.Mutation of P53 may play a role in the transcriptive activation of hTRT gene in non small cell lung cancer.
