甘露醇和生长抑制剂对文心兰离体保存的影响

【摘要】 以文心兰无菌苗为离体保存材料,研究培养基中添加不同浓度甘露醇、矮壮素及脱落酸对文心兰试管苗离体保存的影响。结果表明:在培养温度(24±3)℃,保存前6个月光照强度2 000~2 500 lx,后6个月光照强度500~1 000 lx,光照时间12 h.d-1的培养条件下,MS+6-BA 0.5 mg·L-1+NAA 0.1 mg.L-1+蔗糖30 g·L-1培养基中添加30 g·L-1甘露醇或30 mg·L-1矮壮素(CCC)或2.0 mg·L-1脱落酸(ABA)均能使文心兰试管苗连续保存12个月,从存活率、恢复生长与增殖情况及保存成本等综合考虑,其中以培养基中添加30 g·L-1甘露醇或30 mg·L-1矮壮素处理较适宜文心兰试管苗的保存,保存12个月的存活率达76.7%~80.0%;保存12个月的试管苗在增殖、生根培养基上均能正常恢复生长。

【Abstract】 In vitro preservation of sterile Oncidium plantlets with the addition of mannitol,CCC and/or ABA in the culture medium was investigated.The results showed that(MS + 6-BA) 0.5 mg·L-1 + NAA 0.1 mg·L-1 + sugar 30 g·L-1 in the medium supplemented with mannitol 30 g·L-1 or CCC 30 mg·L-1 or ABA 2.0 mg·L-1 could prolong the plantlet’s shelf life to 12 month at(24±3)℃ under the 12h photoperiod culture condition of 2 000-2 500 lx in the first 6 months,and 500-1 000 lx during the next 6 months.Putting the rates of survival,growth recovery,preservation and proliferation,as well as the cost into consideration,the medium supplemented with 30 g·L-1 mannitol and 30 mg·L-1 CCC was deemed to be most desirable.After 12 months in the medium,the plantlet’s survival rate was 76.7%-80%,and the plant’s growth was normal after cultured in a recovering medium.
