顺铂对卵巢癌细胞几种生长和凋亡相关基因表达的影响
Effects of Cisplatin on the Expression of Several Growth and Apoptosis Related Genes in Ovarian Cancer Cells
【摘要】 顺铂是治疗卵巢癌的一线药物,它与癌细胞内的DNA结合是产生细胞毒性的主要原因.在用基因表达谱芯片筛选顺铂处理IGROV-1细胞产生的差异表达基因基础上,选取其中与细胞生长、凋亡有关的部分重要基因,用实时定量PCR方法进行了差异表达验证.结果表明,顺铂可引起CEACAM1、KLF6、JUN、TP53INP1、MAP2K4、UBE2D3表达上调;导致HERC1、PIK3CA表达下调.
【Abstract】 Cisplatin is one of the primary medicines used in ovarian cancer treatment,it produces cytotoxicity through binding with DNA.Based on the data obtained from gene expression array analysis,some important genes involved in cell growth,apoptosis were selected to validate the differential expression using real-time RT-PCR.The results showed that the expression of six genes was upregulated by cisplatin treatment,these proteins encoded by six genes included CEACAM1,KLF6,JUN,TP53INP1,MAP2K4 and UBE2D3,the expression of two genes was downregulated,these proteins encoded by two genes included HERC1 and PIK3CA.
