人参皂甙与淫羊藿苷复方制剂安全性及抗氧化功能实验研究

【摘要】 目的研究人参皂甙与淫羊藿苷复方制剂作为特殊食品的安全性及其抗氧化作用。方法安全性实验:进行了小鼠急性经口毒性试验、小鼠骨髓嗜多染红细胞微核试验、小鼠精子畸形试验和大鼠30 d喂养试验;抗氧化作用试验采用老龄小鼠作为抗氧化低下动物模型,进行干预性试验,检测各试验组小鼠的血液中MDA含量及GSH-PX活力,测定肝匀浆中SOD活力。结果小鼠急性毒性试验MTD值>20 g/kg bw;人群推荐剂量395倍剂量对小鼠骨髓嗜多染红细胞微核试验、小鼠精子畸形试验结果均为阴性;人群推荐剂量100倍剂量给予大鼠30 d口服未发现临床症状、血液临床检查指标、动物脏器以及病理组织学等指标的改变。与老龄小鼠对照组相比,受试样品中、高剂量组可显著减少老龄小鼠血液MDA含量(P<0.05);中剂量组可显著升高老龄小鼠肝匀浆中SOD活性(P<0.05)以及血液GSH-PX活性(P<0.01)。结论人参皂甙、淫羊藿苷复方制剂具有抗氧化功能,并符合特殊食品安全毒理学要求。

【Abstract】 Objective To test the safety and the effect on anti-oxidation of ginsenoside and epimedium pubescens incariine Astragalus Membranaceus Composite as a kind of health food.Methods Toxicology results were obtained through acute toxicity test,micronucleus test of bone marrow cell,sperm shape abnormality test,and thirty-day feeding tests.Antioxidation indexes were through determination of MDA content,GSH-Px activity and SOD activity.Results ① The amount of 20 g/kg BW(Ginsenoside and epimedium pubescens incariine Astragalus Membranaceus Composite) was fed each time to SPF Kunming mice(female and male),no toxic phenomenon and death were found in animals within two weeks.In the experiment of micronucleus test of bone marrow cell and sperm shape abnormality test in mice,the result was negative.In the thirty-day feeding test,the following dosage was given to the experienced object:0.63 g/kg bw as the low-quantity group,1.26 g/kg bw as the middle,and 2.53 g/kg bw,the highest(25,50,100 times as recommended);After the clinical test,blood test,biochemistry test,organ weight and pharmacology test were performed for the Wistar Rats.No obvious toxic effect was observed.According to the acute toxicity standard rules,no toxic effect was found in the experienced object.② Compared with the control group,the content of MDA in the middle and high dose groups were notably lower(P<0.05),the SOD and GSH-PX activity in the middle dose group significantly increased(P<0.05,P<0.01).Conclusion Ginsenoside and epimedium pubescens incariine Astragalus Membranaceus Composite was effective on antioxidation and could be safely used as a kind of health food.
