大鼠亚慢性镉中毒模型的建立

【摘要】 为建立一种安全、简便和可靠的雄性大鼠亚慢性镉中毒模型,选取30只Wistar雄性大鼠随机分组后,每天分别用含1.25、2.5、5和10 mg/kg剂量的CdCl2溶液灌胃,研究镉离子在不同处理时间对雄性大鼠肝脏和肾脏的损伤情况。结果显示,大鼠血液和肝肾中的镉含量及血清中的尿素浓度与镉离子呈剂量效应关系。染镉56 d后,2.5、5及10 mg/kg处理组大鼠血清中的丙氨酸氨基转移酶活性分别为68.50±11.74、70.67±8.64和72.00±11.67 U/L,显著高于对照组(P<0.05)。病理组织切片结果表明,各处理组中的肝肾组织均出现不同程度的损伤。尽管模型组大鼠有明显的病理学指标变化,然而与对照组相比,镉对动物体重、血清中的乳酸脱氢酶活性以及肌酐含量的影响没有显著性差异。综合所有结果,每天口服浓度为5 mg/kg~10 mg/kg镉溶液且连续处理42 d~56 d,可以建立理想的大鼠亚慢性镉中毒模型。

【Abstract】 To establish a safe,simple and reliable subchronic cadmium poisoning model in rats,30 male Wistar rats were randomly divided into 5 groups.Animals were treated with deionized water containing CdCl2 at 1.25,2.5,5,10 mg/kg by gavage perday,animals in the control group were given deionized water,and the effects of cadmium at different concentrations on liver and kidney was studied.The results were as follows: the contents of cadmium in the serum,liver,kidney and the content of creatinine in serum raised with the dosage’s augmentation and the treatment time’s elongation.The activity of ALT was 68.50±11.74,70.67±8.64 and 72.00±11.67 U/L when the dosage of was 2.5 mg/kg,5 mg/kg and 10 mg/kg perday for 56 days,which was significantly higher than those in the control group(P<0.05).The liver and kidney has obvious pathological lesions,while there was no significant difference on the weights,the creatinine content and LDH activity in serum of rats.The results showed that the establishment of these models treated with different dosages of cadmium(5mg/kg to 10mg/kg) for 42 to 56 days’elongation might be experimentally constructive to the research on the toxicity of cadmium.
