X射线照射致皮质发育障碍模型大鼠致癫性的研究

【摘要】 目的建立X射线照射诱导皮质发育障碍模型,运用快速杏仁核点燃等方法探讨其癫痫易感性等相关特性及测试癫痫易感性的方法。方法 6只SD孕鼠按需要完全随机分为实验组和正常对照组。实验组:在孕16 d采用直线加速器进行X射线照射制作皮质发育障碍模型;正常对照组:不做任何处理。选取P60 d仔鼠,观察一般行为学和大脑外形,HE、N issl染色观察大脑的病理变化;运用水迷宫法测试大鼠学习能力和空间记忆能力;并通过快速杏仁核点燃方法比较两组大鼠发生惊厥所需的刺激次数、后放电阈值。结果与正常对照组比较,实验组大鼠活动性增多,但无自发性癫痫形成;大脑形态发生改变,体积变小,可见明显畸形;HE、N issl染色显示皮层神经元排列紊乱,海马结构紊乱并有异常聚集;在水迷宫实验中,皮质发育障碍组大鼠寻找平台的潜伏期明显长于正常对照组(P<0.05);皮质发育障碍组大鼠的空间探索能力明显低于正常对照组大鼠(P<0.05);快速杏仁核点燃发生所需的刺激次数明显减少[(65.57±10.22)vs(38.57±7.44)次,P<0.01],后放电阈值显著降低[(151.43±51.45)μAvs(74.29±22.25)μA,P<0.05]。结论 X射线照射致皮质发育障碍模型具有癫痫易感性及认知功能障碍,是一种较为理想的皮质发育障碍模型。快速杏仁核点燃是一种较好的测试癫痫易感性的方法。

【Abstract】 Objective To establish the rat model of X-ray irradiation-induced disorder of cortical development,and to investigate its correlation with seizure susceptibility.Methods Six pregnant SD rats were randomly divided into an experimental group and a control group.In the experimental group,the rat model of X-ray irradiation-induced disorder of cortical development was produced on the 16th day of pregnancy with a linear accelerator.The control group was kept without any treatment.The P60d offspring rats were selected to observe the general behavior and the brain shape.The pathological changes of the brain were observed with HE and Nissl staining.The abilities of learning and spatial memory were tested using water maze test.The numbers of simulations and the afterdischarge thresholds of the two groups were compared using rapid electrical amygdala kindling.Results Compared with the control group,the activity of rats in the experimental group increased,yet without developing spontaneous epilepsy.The brain morphology was changed,with smaller volume and obvious malformation.HE and Nissl staining results showed disordered arrangement of cortical neurons,hippocampal structural disorder,and neuron abnormal aggregation.In the water maze test,the latency to find the platform of rats in the experimental group was significantly longer than that in the control group(P<0.05).The space exploration ability of rats in the experimental group was significantly lower than that in the control group(P<0.05).The number of stimulations required to achieve the fully kindled state was significantly reduced in the experimental group(65.57±10.22 vs 38.57±7.44,P<0.01).The afterdischarge threshold was also significantly lowered(151.43±51.45 μA vs 74.29±22.25 μA,P<0.05).Conclusion The rat model of X-ray irradiation-induced disorder of cortical development has obvious seizure susceptibility and cognitive impairment,and can serve as an ideal model of cortical development disorders.Rapid amygdala kindling is a good method to test seizure susceptibility.
