茶多酚与葡萄柚提取物联合应用对变异链球菌生长、产酸的影响
Effect of combined application of tea polyphenols and grapefruit-seed extract on growth and acid production of Streptococcus mutans
【摘要】 目的探讨茶多酚与葡萄柚提取物联合应用对变异链球菌生长、产酸的影响。方法采用棋盘稀释法测定茶多酚葡萄柚提取物混合液对变异链球菌的最低抑菌浓度(MIC)值,计算分级抑制浓度(FIC)指数来判定二者的联合效应;并测定低于MIC浓度的茶多酚葡萄柚提取物混合液对变异链球菌产酸的影响。结果茶多酚和GSE单独用药时,其MIC分别为2 mg/ml和46.88 mg/L,联用时可降低单组分的使用浓度,如茶多酚1 mg/ml联用GSE 37.5 mg/L即可达到联合应用的M IC,FIC=0.75。变异链球菌培养前后的△pH值随混合液浓度的降低而明显增加。结论茶多酚与葡萄柚提取物联合应用对变异链球菌的体外抗菌活性具有相加作用,较各自的单组分用药在更低浓度即可有效抑制变异链球菌的生长、产酸。
【Abstract】 Objective To investigate the effect of combined application of tea polyphenols(TPs) and grapefruit-seed extract(GSE) on the growth and acid production of Streptococcus mutans.Methods Minimal inhibitory concentration(MIC) of mixed solution of TPs and GSE on the growth of Streptococcus mutans(ATCC 25175) was measured by the checkerboard method.Fractional inhibitory concentration(FIC) was calculated accordingly to evaluate the combined effect of TPs and GSE.The effect of mixed solutions of TPs and GSE,with concentrations lower than MIC,on the acid production of Streptococcus mutans ATCC 25175 was also observed.Results Compared with the application of TPs(MIC=2 mg/ml) or GSE(MIC=46.88 mg/L) alone,the combined application of TPs and GSE could lower the MICs of both components.For instance,TPs 1 mg/ml combined with GSE 37.5 mg/L could reach MIC,with an FIC of 0.75;and the △pH value of streptococcus mutans can be increased obviously when the concentration decreased.Conclusion Combined application of TPs and GSE has additive antibacterial effect on Streptococcus mutans.Compared with the application of TPs or GSE alone,combined application of TPs and GSE can effectively inhibit the growth and acid production of Streptococcus mutans at lower concentrations.
