水稻转座子Pong RNAi载体的构建和农杆菌介导的遗传转化
The construction of RNAi vector of a rice Tn pong and Agrobacterium-mediated transformation by regeneration of embryonic callus
【摘要】 为探讨水稻转座子Pong的功能及其对水稻基因组甲基化的影响,构建了水稻转座子Pong的RNAi表达载体TPAⅠ和TPAⅡ,并利用这两个载体采用农杆菌介导法转化水稻(品种松前)的成熟胚愈伤组织,获得PCR结果阳性的再生植株.同时通过不同的农杆菌侵染途径、侵染浓度和侵染时间等,建立并优化了水稻的遗传转化程序.结果表明:农杆菌侵染成熟胚愈伤组织3 min,共培养2~4 d,预培养7 d为最佳的遗传转化方式.
【Abstract】 Two RNAi vectors,TPAⅠ and TPAⅡ,were constructed and the efficient embryonic callus system was established by improving the culture media and time with callus induced from rice mature embryo in this experiment.The results showed that 3 min of infecting time of Agrobacterium,a total of 2~4 days training,pre-trining for seven days and Ka screening concentration of 30 mg/L were the optimizing techniques for the most transgenic procedure in rice.
