5-烯丙基-7-二氟亚甲氧白杨素抑制肝癌细胞NF-κB表达的研究

【摘要】 目的:研究5-烯丙基-7-二氟亚甲氧白杨素(ADFMChR)抑制体外培养人肝癌SMMC-7721细胞增殖和诱导凋亡作用及机制。方法:体外培养SMMC-7721细胞,MTT比色法测定ADFMChR对SMMC-7721细胞增殖活性的影响;Western Blotting法分析ADFMChR对SMMC-7721细胞NF-kB蛋白表达活性的影响。结果:MTT比色法结果显示ADFMChR有效抑制SMMC-7721细胞增殖活性,呈剂量依赖性,其IC50值为3.56μM;Western Blot分析证实ADFMChR下调NF-κB蛋白表达,呈浓度和时间依赖性。结论:ADFMChR具有抑制人肝癌SMMC-7721细胞增殖和诱导细胞凋亡作用,其机制与NF-κB蛋白表达有关。

【Abstract】 Objective:To investigate the effect and mechanism of 5-ally-7-gen-difluormethylene chrysin(ADFMChR),a novel chrysin derivatives,on the proliferation and apoptosis of human liver cancer(SMMC-7721)cell line in vitro.Methods: SMMC-7721cells was cultured in vitro.MTT assay was used to determine the effect of ADFMChR on the proliferation of SMMC-7721cells.Western blot was used to analyze expression of NF-κB protein in SMMC-7721cells.Results:ADFMChR significantly suppressed proliferation of SMMC-7721cell line in a dose-dependant manner.Its IC50was 3.56μM.Western Blotting indicated that expression of NF-κB protein decreased in a dose-and time-dependent manner.Conclusion:ADFMChR could inhibit proliferation and induce apotosis of human liver cancer SMMC-7721cell line.Its mechanism might be associated with down regulation expression of NF-κB protein.
