溶菌酶产生菌的筛选及产酶条件研究
Isolation of high-yielding lysozyme strain and optimization of its fermentation conditions
【摘要】 为了从微生物中获取溶菌酶,本研究采用双层平板法从土壤中筛选获得一株产溶菌酶活性较高的菌株S2-6b。利用摇瓶发酵优化产酶条件,液体发酵培养基成分为可溶性淀粉1.5%,牛肉膏2.5%,玉米浆0.5%,NaCl 0.5%,起始pH值为7.0,培养温度为28℃,接种量为3.0%,摇瓶的装液量为40 mL/300 mL,转速为200r/min。培养40 h酶活力最高,最高酶活力可达6 167 U/mL。
【Abstract】 In order to obtain lysozyme from microorganism,a high-yielding strain for producing lysozyme was screened by double layer plates.The condition for enzyme formation of the strain was studied.The suitable carbon source of medium was soluble starch and the suitable nitrogen source was meat extract and corn steep liquor,whose suitable concentration was 1.5%,2.5% and 0.5% respectively.The optimum temperature and initial pH for enzyme formation were 28 ℃ and pH 7.0 respectively.The inoculum size,liquid volume and shake speed were 3.0%(V/V),40 mL/300 mL 200 r/min respectively.The fermentation cycle was 40 h and the highest lysozyme activity was 6 167 U/mL.
