强碱性壳聚糖分离介质的制备及其性能研究

【摘要】 利用反相悬浮交联法制备了壳聚糖基质,基质再与环氧丙基三甲基氯化铵反应,制得强碱性的壳聚糖阴离子分离介质。研究了分离介质的最佳制备条件,测定了其最大阴离子交换容量和滴定曲线,最后用5′-鸟苷酸对其进行交换性能表征。结果表明,最佳制备工艺为:以1.0%(m/V)的NaOH为催化剂,1.0g壳聚糖基质与5.0mL环氧丙基三甲基氯化铵于50℃反应3.0h,产物为强碱性阴离子分离介质,其最大交换容量为2.88mmol/g干介质,交换吸附结果表明对5′-鸟苷酸最大吸附量为98.59mg/g干介质。

【Abstract】 The chitosan matrix previously made by the methods of inverse phase suspension and chemical cross-linking was then modified by glycidyl trimethylammonium chloride to prepare the strong basic anion separation media.The reaction conditions were optimized,the maximum exchange capacity,pH titration curves and the adsorption of 5′-GMP of the media were studied.The optimal preparation conditions of the media were as followed: under the catalysis of 1.0%(m/V)NaOH,5.0mL glycidyl trimethylammonium chloride reacted with 1.0g chitosan matrix at 50℃ for 3.0h.Under these conditions the maximum exchange was 2.88mmol/g dry media.The results show that it was strong basic anion separation media by the pH titration curves,the study of adsorption indicated that the maximum attachment of 5′-GMP was 98.59mg/g dry media.
