小白菜和菜薹细胞质雄性不育类型的分子鉴定

【摘要】 通过检索NCBI核苷酸数据库获得orf 138和orf 224开放阅读框序列,依据其保守序列设计2对引物,对几种来源不同的小白菜和菜薹细胞质不育系及其保持系进行PCR扩增。orf 138引物在小白菜和菜薹细胞质雄性不育系中均扩增出573 bp的片段,orf 224引物只在一个菜薹雄性不育系中扩增出653 bp的片段。同源性比对发现orf 138引物在雄性不育系中扩增的片断与orf 138基因同源度达100%,表明这几个雄性不育系类型分别为萝卜细胞质不育类型;orf 224引物在雄性不育系中扩增的片断与orf 224基因同源度达100%,表明这一雄性不育系类型为甘蓝型油菜不育类型。

【Abstract】 In order to identify the cytoplasmic male sterile(CMS) types in non-heading Chinese cabbage(Brassica campestris L.ssp.chinensis Makino) and Chinese flowering cabbage(B.campestris L.var.purpurea Baileysh),two pairs primers were designed based on the conserved domains of the sequences of orf 138 and orf 224 genes were got by searching the nucleotide databases of NCBI.The result indicated that a 573 bp specific fragment was produced in all the CMS lines by amplification with the orf 138 primer.The sequence alignment showed that the specific fragment of 573 bp from the CMS lines and the Ogu orf 138 of Ogu CMS in radish reached 100% in homology.Also the specific fragment of 653 bp from the Chinese flowering cabbage CMS line achieved 100% in homology with the Pol orf 224 of Pol CMS in rapeseed(B.napus L.).
