泰山白花丹参提取物对人脐血内皮祖细胞增殖、黏附及分泌NO的影响
Effects of Taishan Salvia miltiorrhiza Bunge.f.alba extract on proliferation,adhesive activity and nitric oxide secretion of umbilical cord blood endothelial progenitor cells
【摘要】 目的:观察单味中药泰山白花丹参提取物(SBE)对人脐血内皮祖细胞(EPCs)增殖、黏附及分泌一氧化氮(NO)的影响。方法:采用密度梯度离心法从脐血获得单个核细胞,将其接种在人纤维连接蛋白包被的培养板上,培养7d后收集贴壁细胞,加入不同浓度SBE继续培养24h;分别采用MTT比色法、黏附能力测定实验和硝酸还原酶法测定EPCs的增殖、黏附和分泌NO能力。结果:与对照组比较,SBE组EPCs增殖、黏附能力提高,培养上清中NO浓度增加(P<0.05),其作用在一定范围内随SBE浓度增加而增强。结论:白花丹参显著改善EPCs功能,这可能是其内皮保护的新机制。
【Abstract】 Objective: To investigate the effects of single Chinese traditional medicine of Taishan Salvia miltiorrhiza Bunge.f.alba extract(SBE) on proliferation,adhesive activit of endothelial progenitor cells(EPCs) and its nitric oxide(NO) secretion.Methods: Total mononuclear cells were isolated from human umbilical cord blood by ficoll density gradient centrifugation and inoculated on the human fibro-ligandin encrusting plate.After 7 days of in vitro culture,adherent cells were collected and incubated with SBE for 24h.The proliferation,adhesive activity and NO secretion of EPCs were assayed using MTT,adhesion determination and nitrate reductase method,respectively.Results: EPCs incubated with SBE showed the capacities higher than those of control in proliferation,adhesion and with a higher NO concentration in the culture supernatant.Conclusion: Salvia miltiorrhiza Bunge.f.alba can improve the function of EPCs,which might be a novel mechanism of its effects in improving vascular endothelial function.
