心脑舒通胶囊对大鼠缺血再灌注脑损伤后细胞骨架的影响

【摘要】 目的:探讨心脑舒通胶囊对大鼠局灶性脑缺血再灌注后细胞骨架的影响。方法:采用线栓法阻塞大脑中动脉致大鼠局灶性脑缺血再灌注模型(MCAO),24h后断头取脑,HE染色观察大鼠皮层区病理形态学改变;免疫组织化学法测定大脑皮层中微管结合蛋白-2(MAP-2)、微管蛋白亚型(β-tublin)的表达。结果:缺血再灌注模型组大脑皮层内皮层区明显水肿,组织结构疏松;MAP-2、β-tublin表达明显降低,与假手术组比较,均有显著性差异(P<0.01)。心脑舒通胶囊能明显减轻皮层区神经细胞水肿;增加缺血侧大脑皮层内MAP-2、β-tublin表达,与模型组比较有统计学意义(P<0.01)。结论:心脑舒通胶囊能够减轻缺血再灌注脑组织损伤程度,保护受损的脑组织,其机制可能与增加脑组织MAP-2、β-tublin表达有关。

【Abstract】 Objective:To study the effect of Xinnaoshutong Capsule on cellular skeleton(MAP-2 and β-tublin) after focal cerebral ischemia and reperfusion in rats.Methods:Rat models of focal cerebral ischemia and reperfusion were established by thread ligation in middle cerebral artery occlusions.After ischemia 1.5 hours and reperfusion 24 hours,the brains were removed to observe the morphological changes of rat cerebral cortical neurons under optical microscope through HE staining,detect the expression of Microtubule-associated Protein-2(MAP-2) and β-tublin in ischemic cerebral brain by immunohistochemistry staining technique.Results:Compared with control group,ischemia 1.5h and reperfusion 24h model group had obviously neuronal edema and structure loosing in cerebral cortex.The expression of MAP-2 and β-tublin were significantly decreased(P<0.01).Xinnaoshutong Capsule group could allevate neuronal edema,increase the expression of MAP-2 and β-tublin in brain tissue(P<0.01).Conclusion:Xinnaoshutong Capsule had the protective effects in lightening injury degree of brain tissues after cerebral ischemia and reperfusion.The effect may be related to the increase of MAP-2 and β-tublin expression in brain tissue.
