宣肺中药对COPD模型大鼠肺组织NKA及VIP含量的影响

【摘要】 目的:观察宣肺中药(加味桔梗汤)对慢性阻塞性肺疾病(COPD)模型大鼠肺组织VIP、NKA的影响,从神经肽角度探讨其作用机制。方法:用随机数字表法将40只Wister大鼠分为为正常组、模型组、模型给药组及正常给药组;采用气管滴注脂多糖加熏香烟联合造模方法建立COPD大鼠模型。正常组、模型组灌胃蒸馏水,各给药组于第2-13d、15-28d以加味桔梗汤(桔梗、生甘草、紫苑、浙贝)灌胃。第29d处死大鼠,测定其肺泡灌洗液(BALF)中TNF-α、IL-1β含量及细胞分类计数,并检测其肺组织中VIP、NKA含量。结果与正常组相比,模型组大鼠BALF中TNF-α、IL-1β含量及淋巴细胞计数均明显增高,同时伴有肺组织NKA含量明显升高,VIP含量有减少趋势;经加味桔梗汤干预后,BALF中TNF-α含量及淋巴细胞计数均明显降低,IL-1β含量表现出下降趋势,且肺组织中NKA含量显著降低,VIP含量虽有增高趋势;相关分析显示,肺组织中NKA、VIP含量变化与IL-1β含量及淋巴细胞计数变化显著相关。结论:加味桔梗汤对COPD模型大鼠气道炎症的改善作用,可能与调节肺内神经肽NKA、VIP含量有关。

【Abstract】 Objective:To observe the influence of facilitating lung on contents of NKA and VIP in lung tissues in rats with COPD,and to investigate the mechanism.Methods:All rats were randomly divided into normal control group,model group,treating group and normal treating group.The rat model of COPD was established by using the method of cigarette smoking combined with intratracheal injection of LPS.The normal control group,model group were given intragastrically distilled water,and other groups were given the modified Jiegeng Decoction intragastrically except 14th day and 28th day.Detecting contents of TNF-α,IL-1β and counting lymphocyte in BALF,and checking out the content of NKA and VIP in lung tissues.Results:The contents of TNF-α,IL-1β,lymphocyte and NKA were markedly higher,while VIP was lower,in the model group,compared with those in the normal control group.Compared with model group,the contents of TNF-α,IL-1β,lymphocyte and NKA were significantly decreased in treating group,but the content of VIP improved further in treating group.Conclusion:The modified Jiegeng Decoction can regulate the contents of NKA and VIP to ameliorate the inflammation of respiratory tract in rats with COPD.
