天门冬对慢性辐射损伤大鼠心脏的保护作用及其机制

【摘要】 目的:研究天门冬水煎剂对慢性辐射损伤大鼠心脏的保护作用及其机制。方法:SD大鼠以200mGy照射,照射50次建立慢性辐射损伤模型,以天门冬水煎剂进行治疗;采用硫代巴比妥酸和黄嘌呤氧化法分别检测血清和心脏中丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性;应用Real-time PCR和免疫印记法检测各组心脏组织中c-myc的表达。结果:天门冬水煎剂治疗组与模型组相比,血清与心脏中SOD的活性均明显增高、MDA含量显著降低(P<0.05),呈剂量依赖性;与模型组相比,天门冬能够使大鼠心脏组织内的c-myc表达下降(P<0.05)。结论:天门冬可以减轻慢性辐射对心脏的损伤作用,其机制可能与降低心脏c-myc基因的表达有关。

【Abstract】 Objective: To study the protective Effect and mechanism of Chinese Asparagus on damaged rat’s heart induced by chronic radiation.Methods: SD rats were irradiated by 200 mGy×50 times,the treatment group were administered by gavage with different dose of the Chinese Asparagus.Activity of SOD was measured by SOD kit with thiobarbituric acid method and the contents of MDA were measured by xanthine oxidase method.The expression of c-myc mRNA and c-myc protein of the heart were detected by Real-time PCR and Western-blot.Results: Compared with control group,the Chinese Asparagus can increased the activity of SOD and decreased the contents of MDA(P<0.05).The Chinese Asparagus can effectively decreased the expression of c-myc mRNA and protein significantly(P<0.05).Conclusion: The protective mechanism of Chinese Asparagus on the heart of chronic radiation damage may be relevant to the decreased expression of c-myc.
