洋桔梗组培技术研究

【摘要】 [目的]探讨外源激素水平等对洋桔梗愈伤诱导、不定芽分化、继代增殖、生根的影响,以建立其组织培养工厂化育苗生产体系。[方法]通过无菌插种获得无菌苗,取其带腋芽茎段为外植体,采用随机区组试验筛选愈伤形成、不定芽分化、增殖与不定根形成的适宜激素水平。[结果]适宜洋桔梗愈伤与不定芽分化的培养基为MS+6-BA 0.1~0.5 mg/L+NAA 0.1~0.5 mg/L;继代增殖适宜培养基为MS+6-BA 0.1~0.5 mg/L+NAA 0.1~0.5 mg/L+GA 0.1~0.5 mg/L+干酪素50~100 mg/L;生根适宜的培养基为1/2MS+NAA 0.1~0.3 mg/L或IBA 0.5 mg/L或IAA 0.5 mg/L,25 d内生根率可达95%以上;在蛭石∶珍珠岩∶腐叶土=4∶4∶2基质中驯苗,成活率可达96%以上。[结论]利用组织培养技术对洋桔梗进行快速繁殖,实现其工厂化育苗具有重要的现实意义。

【Abstract】 [Objective] The aim was to discuss the effects of external hormone on callus induction,uncertain roots differentiation,proliferation and adventitious roots formation of E.grandiforum,to establish manufacturing system of raising seedlings in industrial scale.[Method] Obtaining aseptic plant in the way of aseptic seeding,applying parts of the plant stems as explants,then using randomized experiments to screen the best hormone level for callus formation,uncertain roots differentiation,proliferation and adventitious root formation.[Result]The results show that the suitable culture medium for callus and uncertain roots differentiation of E.grandiforum was MS+BA 0.1-0.5 mg/L+NAA 0.1-0.5 mg/L,the suitable culture medium for proliferation was MS+6-BA 0.1-0.5 mg/L+NAA 0.1-0.5 mg/L+GA 0.1-0.5 mg/L+50-100 mg/L casein,the suitable culture medium root formation was 1/2MS+NAA 0.1-0.3 mg/L or IBA 0.5 mg/L or IAA 0.5 mg/L,which makes the rooting rate reach 97% during 25 days;if the sprouts are transplanted into the substrate in which the rate among ermiculite,perlite and humus soil is 4∶4∶2,the survival can reach over 96%.[Conclusion] Tissue culture technology is used in rapid propagation of E.grandiforum,which has realistic significance for raising seedlings in industrial scale.
