粗糙链孢霉杂交试验的新方法

【摘要】 [目的]改进粗糙链孢霉杂交试验的方法。[方法]对粗糙链孢霉培养条件、杂交方法、最佳取材时间及子囊孢子成熟时对应的子囊果的形状进行研究。[结果]粗糙链孢霉菌种的活化、杂交均可在玉米培养基上培养;在杂交时,将菌丝挑到无菌水中制成菌液,用菌液接种,方便快捷;在25℃,21 d左右取黑色长刺状凸起的子囊果成熟度好,压片效果好。[结论]试验结果可应用于遗传学教学实践。

【Abstract】 [Objective] The aim of the study was to modify the cross experiment of Neurospore crassa.[Method] Cultivation conditions and the proper shape of the ascocarp,especially some detailed experiment steps were studied.[Result]The study showed that in the corn culture medium,the activation and the cross experiment of Neurospore crassa grew well;at the time of cross experiment,bacteria liquid were prepared by using hypha in sterile water,the inoculation of bacteria liquid was convenient and quick.After cultivating 21 d under 25 ℃,the ascuses began to mature.The characters of mature ascocarps were black and long spinous.[Conclusion]The experimental results were applicable to the practice of genetics experimental teaching.
