Hg2+胁迫对小麦幼苗生长及其过氧化物酶·淀粉酶活性的影响
Effects of Mercury Stress on Wheat Growth,POD and Amylase Activity
【摘要】 [目的]研究Hg2+对小麦种子萌发和幼苗生长的影响。[方法]以郑州9023为供试材料,采用室内水培方法,研究不同浓度(0.025、0.050、0.100、0.200、0.300、0.400、0.500 mmol/L)Hg2+溶液浸种处理后,对小麦发芽率及幼苗苗高、根长、苗鲜重及叶片、根和萌发种胚中过氧化物酶(POD)、淀粉酶活性等的影响。[结果]低浓度Hg2+(≤0.100 mmol/L)处理对小麦种子萌发、幼苗苗高、根长和苗鲜重的影响较小;在Hg2+浓度>0.100 mmol/L时,对小麦种子萌发和幼苗的正常生长有明显抑制作用。低浓度Hg2+(≥0.025 mmol/L)可诱导小麦幼苗叶片、根和萌发种胚中POD活性明显升高,而抑制淀粉酶活性使之明显下降,并且这种影响随着Hg2+浓度的增大而增强。[结论]Hg2+胁迫可以改变小麦幼苗叶片、根和萌发种胚中POD和淀粉酶活性,从而影响其膜脂氧化正常代谢,导致生长所需的能量和底物的供应不足,最终抑制了幼苗的生长。
【Abstract】 [Objective] The aim was to study the effect of mercury stress on seed germination and seedlings growth.[Method] Zhengzhou 9023 was used as the experimental material and cultured in water.After the seeds were soaked in different concentrations of Hg2+(0.025,0.050,0.100,0.200,0.300,0.400 and 0.500 mmol/L),the germination rate and seedling height,root length,seedling’s fresh weight,and the activities of peroxidase(POD) and amylase in leaf,root and germinating embryo were measured.[Result] Low concentrations of Hg2+(≤0.100 mmol/L) had little effect on seed germination,seedling height,root length and fresh weight;high concentrations of Hg2+(>0.10 mmol/L) could significantly inhibit seed germination and seedling growth.Low concentration of Hg2+(≥ 0.025 mmol/L) could significantly increase POD activity and inhibit the amylase activity,and the effects increased with the increasing of Hg2+concentrations.[Conclusion] Hg2+ stress could change the activities of POD and amylase in leaf,root and germinating embryo,thus influence the energy and substrate supply which was required for normal metabolism of lipid oxidation,and inhibit seedling growth ultimately.
