一株高效秸秆纤维素降解真菌的分离、鉴定及系统发育分析

【摘要】 通过刚果红染色法初筛,结合产酶活性测定复筛,从奶牛、山羊瘤胃内容物中分离到一株纤维素高效降解菌NL-3,克隆了该菌的18S rDNA序列,并以其同源性为基础构建了相关属种的系统发育树。结果表明,18S rDNA序列全长504 bp,Blast显示该菌株与青霉菌属同源;NL-3菌株在进化关系上也与青霉菌属(Penicillium)聚成一族。特别是与Penicillium decumbensstrain JU-A10的同源性最高,序列相似性达99%。结合菌株形态学及18S rDNA基因序列分析结果对菌株进行鉴定,初步鉴定为青霉属的斜卧青霉(Penicillium decumbens)。

【Abstract】 A fungal strain named as NL-3 that could degrade cellulose was isolated from the rumen of cows and sheep.The 18S rDNA gene sequence for strain NL-3 was amplified and sequenced.The results showed that the complete 18S rDNA gene sequence was 504 bp.A phylogenetic tree was constructed by comparing the 18S rDNA sequence with other relative fungus species in the GenBank database.The strain NL-3 and Penicillium decumbens strain JU-A10 constituted a branch with the similarity value as 99%.According to the morphological characteristics and phylogenetic analysis,the strain NL-3 was identified as Penicillium decumbens.
