【摘要】 目的探讨补肾活血方防治系膜增生性肾小球肾炎(MsPGN)大鼠肾小球硬化的作用机制。方法将54只雄性SD大鼠随机分为正常组、模型组、治疗组,每组18只,尾静脉注射抗大鼠胸腺细胞抗血清(ATS)建立MsPGN模型,治疗组在ATS注射后即以补肾活血方灌胃,每日1次,灌胃剂量为5ml/kg。各组分别于第2、4、8周各取6只大鼠,测定尿蛋白(Upro)、血尿素氮(BUN)和肌酐(SCr)含量;切取肾脏用于病理学观察及免疫组化检测肾组织型胶原(Col-Ⅳ)和层粘连蛋白(LN)的表达。结果病理学检查治疗组较同时间点模型组相比,肾小球系膜细胞增生程度明显减轻,系膜基质明显减少。治疗组大鼠各时间点Upro、BUN和SCr含量较模型组均明显降低(P<0.05或P<0.01),治疗组8周时,肾组织LN、Col-相对含量、积分光密度(ILD)值均较模型组亦明显降低(P<0.05)。结论补肾活血方能抑制MsPGN大鼠肾组织Col-Ⅳ和LN的表达,这可能是防治肾小球硬化机制之一。更多还原

【Abstract】 Objective To investigate the mechanism of Bushen Huoxue Recipe (Recipe to reinforce the kidney and activate blood) in the treatment of glomerulosclerosis of the rats with mesangial proliferative glomerulonephritis (MsPGN).Methods Fifty-four SD male rats were randomized into normal group, model group, and treated group with 18 rats in each. The MsPGN models were made by injection of anti-rat-thymocyte antiserum (ATS) through the tail vein. After modeling, the treated group was administered Bushen Huoxue Recipe with a dosage of 5ml/kg, once a day. Six rats from each group were selected in the second, fourth, and eighth week respectively to determine the content of urine protein (Upro), blood urea nitrogen (BUN), and serum creatinine (SCr), and the kidney was cut for pathological observation and immunohistochemical detection of the kidney tissues, the expression of Ⅳ-type collagen (Col-Ⅳ) and laminin (LN).Results The pathological determination showed that, at the same time points, the hyperplasia of mesangial cells in the treated group was significantly relieved and mesangial matrix reduced as compared with the model group. The content of Upro, BUN and SCr at all time points in the treated group was significantly decreased as compared with the model group (P<0.05 or P<0.01). In the eighth week, the content of LN and Col-Ⅳ of the kidney tissue and the value of integral optical density (ILD) in the treated group were both significantly lowered as compared with the model group (P<0.05).Conclusion Bushen Huoxue Recipe inhibits the expression of Ⅳ-type Collagen and Laminin of the MsPGN rats, being the possible mechanism in the treatment of glomerulosclerosis.更多还原
