魔芋葡甘聚糖季铵盐接枝共聚物的合成及生物学活性

【摘要】 以硝酸铈铵为引发剂,以魔芋葡甘聚糖作载体,用甲基丙烯酰氧乙基-辛烷基-二甲基溴化铵为活性单体合成了魔芋葡甘聚糖季铵盐接枝共聚物.用红外光谱、核磁共振、差示扫描量热分析仪和激光粒度Zeta电位仪对接枝产物进行了表征和测定,用悬菌定量杀菌实验研究了其抗菌性能.结果表明:甲基丙烯酰氧乙基-辛烷基-二甲基溴化铵季铵盐单体通过其双键接枝到魔芋葡甘聚糖上;魔芋葡甘聚糖的零电位pH=4.5,而魔芋葡甘聚糖季铵盐接枝共聚物的零电位pH=10.2,由于阳离子季铵基团的引入使魔芋葡甘聚糖的零电位发生了较大的位移;所合成的魔芋葡甘聚糖季铵盐接枝共聚物对大肠杆菌、金黄色葡萄球菌与白色念珠菌在振荡作用15 min后,平均杀菌率分别为99.99%、99.99%和92.55%.

【Abstract】 Methacryloxyethyl-octyl-dimethyl ammonium bromide was grafted onto konjac glucomannan(KGM) carrier by using ceric ammonium nitrate as the initiator to obtain a graft copolymer.The graft copolymer was represented and determined by hydrogen nuclear magnetic resonance spectroscopy(1H NMR),Fourier transform infrared spectroscopy(FTIR),differential scanning calorimeter(DSC),a laser particle sizer and a zeta potential analyzer.The antimicrobial properties of the graft copolymer were studied by the suspension quantitative test.The results show that: methacryloxyethyl-octyl-dimethyl ammonium bromide monomer can be grafted onto the KGM through the double bond thereof;the zero potential points of the KGM and the graft copolymer are pH 4.5 and pH 10.2,respectively;the large shift of the zero potential point of the KGM occurs due to the introduction of the cationic quaternary ammonium groups;and the KGM graft copolymer has significant effect on inhibiting E.coil(8099),S.aureus(ATCC 6538) and C.albicans(ATCC10231) after being shaken for 15 min,and the average bactericidal rate are respectively 99.99%,99.99% and 92.55%.
