绿茶多酚对慢性脑低灌注大鼠皮质抗氧化能力的影响
The effect of green tea polyphenols on the antioxidant ability after chronic cerebral hypoperfusion in rats
【摘要】 目的:探讨不同剂量的绿茶多酚对慢性脑低灌注大鼠抗氧化能力的影响。方法:健康雄性Wist-ar大鼠随机分为假手术组、2VO加生理盐水组、2VO加GTP100组(绿茶多酚100mg·kg-1·d-1)、2VO加GTP400组(绿茶多酚400mg·kg-1·d-1)。采用大鼠双侧颈总动脉结扎(2VO)制备慢性脑低灌注模型,结扎后从第4周开始给药,给药4周后采用生化方法检测脑皮质的抑制羟自由基能力和SOD的活力,并采用实时定量PCR方法检测皮质的Nrf2mRNA的表达。结果:绿茶多酚增强SOD活性和抑制羟自由基能力,增加皮质Nrf2mRNA的表达。400mg·kg-1·d-1剂量的绿茶多酚的作用更显著。结论:绿茶多酚增强慢性脑低灌注大鼠脑组织的抗氧化能力,尤其400mg·kg-1·d-1剂量的绿茶多酚作用更明显。
【Abstract】 Objective:To investigate the effect of green tea polyphenols on the antioxidant ability after chronic cerebral hypoperfusion in rats.Method:Healthy male Wistar rats were used for the study.All the rats were randomly divided into 4groups:sham-operated group,2VO加normal saline group,2VO加 GTP100group (green tea polyphenols 100mg·kg-1·d-1),2VO加GTP400group(green tea polyphenols 400mg·kg-1· d-1).The chronic cerebral hypoperfusion models were made by permanent bilateral occlusion of common carotid arteries(2VO).After model was establised,drugs were administered from the fourth week.The capability of inhibiting hydroxy radical and the activity of superoxide dismutase(SOD)in the cortex were detected bio-chemically after the drug were given for 4weeks.And the levels of Nrf2 mRNA were examined by real-time quantitatie polymerase chain reaction.Result:Green tea polyphenols enhanced the capability of inhibiting hy-droxy radical and the activity of superoxide dismutase,up-regulated the mRNA levels of Nrf2.To be treated with 400mg·kg-1·d-1 green tea polyphenols had the significant effect.Conclusion:Green tea polyphenols enhanced the antioxidant mechanism after chronic cerebral hypoperfusion.The dosage(400mg·kg-1·d-1) of green tea polyphenols had the significant effects.
