大鼠成骨细胞RANKL-OPG mRNA表达的增龄性变化及淫羊藿甙的干预作用
Changes with age and effect of Icariine on genetic expression of RANKL-OPG in Rat’s Osteoblasts
【摘要】 目的:观察体外培养大鼠成骨细胞RANKL-OPG mRNA的表达随年龄增长的变化,及淫羊藿甙的干预作用,探讨RANKL-RANK-OPG系统在骨质疏松发病中的作用及淫羊藿甙有效防治骨质疏松的机理。方法:取出生24 h、3月龄、6月龄和12月龄SD雌雄大鼠的成骨细胞进行体外培养,MTT测定淫羊藿甙促成骨细胞增殖的最佳浓度,干预后RT-PCR检测RANKL-OPGmRNA的表达。结果:RANKL-OPG的比例随年龄增长逐渐变大,而淫羊藿甙可以一定程度上改变上述变化趋势。结论:RANKL-OPG比例的变化在骨质疏松发病中起到重要作用,淫羊藿甙通过调节RANKL-OPG比例的增龄性变化延缓了骨丢失随年龄增长而加剧的趋势,从而有效地防治骨质疏松症。
【Abstract】 Objective:To observate the effect of age and icariine on genetic expression of RANKL-OPG in OB,so that to probe into role of RANKL-RANK-OPG in Pathogenic Mechanism of Osteoporosis and the detailed mechanism of prevention and cure of osteoporosis with icariine.Methods:Establishing the culture in vitro system of Osteoblasts(OB) from calvaria of SD rats that newborn,3 months,6 months and 12 months.Cell proliferation of OB cultured in differenent concentration icariine was measured by MTT Method,and the optimization concentration of the serum containing JGG would be obtained.Then culture the cells with the serum containing icariine.The levels of RANKL and OPG mRNA were measured by SYBR GREEN real-time quantitative reverse transcription polymerase chain reaction(RT-PCR).Rusults:It was found that genetic expression of RANKL in OB increased with aging,while OPG decreased,and it can be changed by the icariine.Conclusions:The proportion of RANKL-OPG in OB is an important role in Pathogenic Mechanism of Osteoporosis.The detailed mechanism of prevention and cure of osteoporosis with icariine maybe is the physiological changes of RANKL-OPG,and the bone loss is decelerated effectively.
