激活素A对新生大鼠缺血缺氧后脑组织巢蛋白表达的影响

【摘要】 目的探讨激活素A对新生大鼠缺氧缺血性脑损伤(HIBD)脑组织巢蛋白的表达变化。方法将7日龄Wistar大鼠120只随机分为3组,假手术组、缺氧缺血性脑损伤(HIBD)模型组和激活素A治疗组,每组各40只。每组根据处死的时间不同又分为5个亚组:6h组、24h组、3d组、7d组、14d组,每亚组各8只。HE染色观察神经细胞形态变化,采用免疫组化的方法测定巢蛋白的表达。结果HIBD组缺氧缺血侧大脑组织巢蛋白活性逐渐增高,3d显著增高,7d达高蜂,然后逐渐下降,14d仍有少量表达;与HIBD模型组各时间点相比,激活素A治疗组各时间点脑组织巢蛋白表达明显增高(P<0.01)。结论激活素A治疗增高缺氧缺血后巢蛋白的表达,对缺氧缺血性脑损伤有保护作用。

【Abstract】 Objective To explore the effect of activin A on nestin expression in neonatal rats with hypoxic-ischemic brain damage(HIBD).Methods One hundred and twenty 7-day-old Wistar rats were divided into three groups randomly(n=40 in each group):Shamoperation group,HIBD hypoxic-ischemic brain damage group and activin A group,and each group were further divided into five subgroups(n=8 in each subgroup)based on different time points after HIBD(6 h,24 h,3 d,7 d,14 d).The morphologic change of nerve cell was observed by HE staining,and the expression of nestin was determined by immunohistochemical method.Results Compared with sham-operation group,the expression of nestin was increased in HIBD group(P<0.01).The nestin expression was higher significantly at 72 h,it reached the peak at 7 d,and then decreased gradually.There was still a small amount of expression at 14 d.The expression of nestin in activin A group was higher significantly at each time point than that in HIBD group(P<0.01).Conclusion Activin A could increase the expression of nestin,protect the hypoxic-ischemic brain and reduce injury after hypoxia-ischemia.
