大黄■虫丸对球囊损伤内皮诱导的家兔动脉粥样硬化的影响
Effect of Dahuang Zhechong Pill on the experimental atherosclerosis induced by balloon angioplasty in rabbits
【摘要】 目的:观察大黄■虫丸对球囊损伤内皮诱导的家兔动脉粥样硬化的影响。方法:采用机械损伤动脉内皮合并高脂饲料喂养的方法建立家兔动脉粥样硬化模型。雄性新西兰家兔分为正常对照组,假手术组,模型对照组,阳性对照组,大黄■虫丸低、高剂量组。正常对照组、假手术组与模型对照组给予蒸馏水,阳性对照组给予丹参0.5g/kg,大黄■虫丸低、高剂量组分别给予大黄■虫丸0.9、1.8g/kg。灌胃给药,每天1次,连续60d。常规检查血管璧的组织病理形态和超微结构,采用图像分析系统测定血管壁内膜厚度(IT)和内-中膜厚度(IMT),用分光光度法测定血管壁羟脯氨酸含量。结果:大黄■虫丸能减小动脉粥样硬化斑块面积,减少血管平滑肌细胞增殖和胶原纤维增生,抑制内膜增厚,减轻血管平滑肌细胞线粒体肿胀,减少泡沫样细胞形成;降低IT、IMT值,降低血管壁羟脯氨酸含量。结论:大黄■虫丸有抗动脉粥样硬化作用,机制可能与其疏通经络、活血化瘀、破於生新等功效有关。
【Abstract】 Objective: To study the effects of Dahuang Zhechong Pill(DHZCP) on the experimental atherosclerosis induced balloon angioplasty in rabbits.Methods: Atherosclerosis model was established by combination of balloon angioplasty-induced endothelial injury and high cholesterol feeding in rabbit.Male New Zealand rabbits were divided into six groups in random: normal control,sham,model,positive control and two doses of DHZCP-treated groups.Rabbits in two doses of DHZCP-treated groups were intragastrically administered 0.9 and 1.8 g/kg DHZCP for 60 days respectively,and rabbits in positive control group were given 0.5 g/kg Danshen in the identical fashion.Morphology and ultrastructure of the aorta wall were observed by the routine methods.Intima thickness(IT) and intima-media thickness(IMT) of aorta were examined with the image analyzing system.Hydroxyproline level in the vessel wall was spectrophotometerically measured.Results: DHZCP significantly lessened the atherosclerotic plaque area,inhibited the proliferation of vascular smooth muscle cells and collagen to diminish pachysis of vascular intima,relieved swelling of mitchondria in vascular smooth muscle cells,and formation of foam cells in vascular intima.DHZCP also reduced IT,IMT,and decreased hydroxyproline content.Conclusion: DHZCP is able to produce a significant anti-atherosclerotic action.
