芪棱汤对脑缺血再灌注损伤大鼠血脑屏障通透性及基质金属白酶-3表达的影响

【摘要】 目的观察芪棱汤对脑缺血再灌注损伤大鼠血脑屏障通透性及基质金属蛋白酶-3(MMP-3)表达的影响。方法将大鼠随机分为假手术组、模型组、芪棱汤去养阴药组(对照组)及芪棱汤组(治疗组)。采用颈内动脉线栓法复制大鼠脑缺血再灌注(MCAO/Reperfusion)模型。通过测定渗出血管外的伊蓝(EB)含量来评价血脑屏障通透性的变化,在再灌注不同时点采用免疫组织化学法检测MMP-3的表达。结果在再灌注各时间点,治疗组、对照组与模型组比较均有显著差异,治疗组、对照组EB渗出量、MMP-3表达少于模型组;治疗组与对照组有显著差异,治疗组EB渗出量、MMP-3表达少于对照组。结论芪棱汤对脑缺血再灌注后的保护作用,其机制可能与通过抑制MMP-3表达保护血脑屏障有关。

【Abstract】 Objective:To study the effect of Qileng Decoction on the blood-brain barrier permeability and the MMP-3 expression in rats with cerebral ischemia reperfusion. Methods:Rats were randomly divided into the sham-operation group,the model group,the control group and the treatment group. The control group was given Qileng Decoction without nourishing yin herb. The treatment group was used Qileng Decoction. The MCAO model was established by the method of internal carotid artery occlusion by suture. The variation of the blood-brain barrier permeability was detected by the EB content exudation vascular. The MMP-3 expressions in different time points were detected by the immunohistochemistry method. Results:In different reperfusion time points,there were some signif-icant differences in the treatment group,the control group and the model group. The EB exudation content and the MMP-3 expression of the treatment group and the control group were lower obviously than those of the model group. There was a remarkable difference between the treatment group and the control group. The EB exudation content and the MMP-3 expression of the treatment group were lower significantly than those of the control group. Conclusion:The mechanism of that Qileng Decoction could protect the function after cerebral ischemia reperfusion could be related to its inhibition the MMP-3 expression to protect the blood-brain barrier.
