小檗碱对氧化型低密度脂蛋白诱导的内皮细胞损伤的保护作用

【摘要】 目的探讨小檗碱是否具有保护内皮细胞的作用。方法用不同浓度的小檗碱预处理人脐静脉内皮细胞(HUVECs),再用400mg/L的氧化型低密度脂蛋白(OX-LDL)诱导HUVECs损伤,通过评价细胞增殖、上清液中一氧化氮(NO)及内皮素(ET-1)含量来评价小檗碱对内皮细胞的保护作用。结果 400mg/LOX-LDL能明显抑制HUVECs增殖,下调细胞分泌NO含量,上调ET-1含量;而与模型组相比,小檗碱预处理组细胞增殖活性明显增加,NO含量增加,ET-1含量降低,但随着小檗碱浓度的增加,这种保护作用逐渐减弱。结论小檗碱能够减轻OX-LDL对HUVECs的损伤,对HUVECS具有一定的保护作用。

【Abstract】 Objective:To explore the protective effects of berberine on endothelial cells.Methods:Human umbilical vascular endothelial cell(HUVEC) were pretreated with different concentrations of berberine.Then,the HUVEC injury was induced by the OX-LDL.The protective effect was measured by the contents of the cell proliferation,the NO content and the ET-1 level.Results:400mg/L OX-LDL could inhibit HUVEC growth,decrease NO content and increase the ET-1 content.Compared with the model group,the proliferation activity was increased obviously in the berberine pretreatment group.Meanwhile,the NO content was increase and the ET-1 level was decreased.But this protection effect was reduced while the berberine level promotion.Conclusion:Berberine can relieve the HUVEC injury induced by OX-LDL.It can protect the HUVEC.
